Current insights on therapeutic and diagnostic approaches based on molecular pathogenesis for Alzheimer’s disease

Alzheimers disease (AD) is an age associated neurodegenerative disease and memory deficit is a major symptom for patients. As pathological hallmarks in their brains, extracellularly deposited amyloid plaques which are mainly composed of beta amyloid peptides, intracellularly accumulated neurofibrillary tangles with abnormally hyperphosphorylated tau protein and neuronal loss are well described. Brain inflammation is also known as strong pathological factors to exacerbate brain pathologies. Various cell types including neuron, microglia and astrocyte are tightly associated with brain pathologies in AD. To study AD pathogenesis, several animal models are reported such as APP/PS1 and tau transgenic mice and APP and tau knock-in mice. These animal model systems give us to the opportunity for therapeutic target identification and validation for the novel target. Recently, AD patients-derived IPSC and brain organoid models are available. It opens new era to be possible personalized medicine for development of targeted drug screening based on patient-specific genetic and other factors. Since there is no FDA approved disease-modifying drug for AD, it is good time to challenge new paradigm for drug design and application for AD therapy. In the present seminar, current trends of AD basic research and their applications are discussed in detail.
